Adalimumab (Humira®)

The drug Adalimumab (trade name Humira®) is a recombinant human IgG1 monoclonal antibody specific for human tumor necrosis factor alpha (TNF-α). Adalimumab was created using phage display technology resulting in an antibody with human derived heavy and light chain variable regions and human IgG1:kappa constant regions. Adalimumab is produced by recombinant DNA technology in a mammalian cell expression system and it consists of 1330 amino acids and has a molecular weight of approximately 148 kilodaltons. Adalimumab binds specifically to (TNF-α) and blocks its interaction with the p55 and p75 cell surface TNF receptors. Adalimumab also lyses surface TNF expressing cells in vitro in the presence of complement. Identification of biomarkers for (non-)response and risk factors for adverse drug reactions that might be related to serum concentrations and maintaining the effective concentration of Adalimumab in order to potentially avoid some side effects with a reliable method might be beneficial. Enzyme immunoassay (ELISA) test kit for the determination of free Adalimumab (Humira®) in serum and plasma. The catcher is TNF-α and the detection antibody is a HRP-labeled mAb binding at the Fc part of the drug. Due to the assay design there is full cross reactivity with other TNF-α catchers. This kit is for research use only, not for use in diagnostic procedures.

Intended use

This ELISA kit applies to the in vitro quantitative determination of Adalimumab (Humira®) concentrations in serum, plasma and other biological fluids.

Test principle

This ELISA kit uses the Sandwich-ELISA principle. The micro ELISA plate provided in this kit has been pre-coated with an antigen specific to Adalimumab (Humira®). Standards or samples are added to the micro ELISA plate wells and combined with the specific antigen. Then a biotinylated detection antigen specific for Adalimumab (Humira®) and Avidin-Horseradish Peroxidase (HRP) conjugate are added successively to each micro plate well and incubated. Free components are washed away. The substrate solution is added to each well. Only those wells that contain Adalimumab (Humira®), biotinylated detection antigen and Avidin-HRP conjugate will appear blue in color. The enzyme-substrate reaction is terminated by the addition of stop solution and the color turns yellow. The optical density (OD) is measured spectrophotometrically at a wavelength of 450 nm ± 2 nm. The OD value is proportional to the concentration of Adalimumab (Humira®). You can calculate the concentration of Adalimumab (Humira®) in the samples by comparing the OD of the samples to the standard curve.

Typical data

As the OD values of the standard curve may vary according to the conditions of the actual assay performance (e.g. operator, pipetting technique, washing technique or temperature effects), the operator should establish a standard curve for each test. Typical standard curve and data is provided below for reference only.

Precision

Intra-assay Precision (Precision within an assay):
3 samples with low, middle, high concentration of Adalimumab (Humira®). And the sample of Adalimumab (Humira®) were tested 30 times on one plate, respectively.
Inter-assay Precision (Precision between assays):
3 samples with low, middle, high concentration of Adalimumab (Humira®). And the sample of Adalimumab (Humira®) were tested on 3 different plates, 30 replicates in each plate.

Recovery

Matrices listed below were spiked with certain level of Adalimumab (Humira®) and the recovery rates were calculated by comparing the measured value to the expected amount of Adalimumab (Humira®) in samples.

Matrix Recovery range (%) Average Recovery (%)
Serum (n=5) 88-105 97
EDTA plasma (n=5) 88-100 95
heparin plasma (n=5) 91-98 94

Linearity

The linearity of the kit was assayed by testing samples spiked with appropriate concentration of Adalimumab (Humira®) and their serial dilutions. The results were demonstrated by the percentage of calculated concentration to the expected.

Serum (n=5) EDTA plasma (n=5) heparin plasma (n=5)
1:2 Range (%) 88-105 92-103 94-100
Average (%) 93 93 94
1:4 Range (%) 91-99 88-102 92-101
Average (%) 95 95 94
1:8 Range (%) 89-106 90-106 93-99
Average (%) 93 93 94
1:16 Range (%) 94-99 93-102 88-104
Average (%) 95 95 94

Stability

The stability of Adalimumab (Humira®) is determined by the loss rate of activity. The loss rate of Adalimumab (Humira®) is less than 5% within the expiration date under appropriate storage condition. To minimize extra influence on the performance, operation procedures and lab conditions, especially room temperature, air humidity, incubator temperature should be strictly controlled. It is also strongly suggested that the whole assay is performed by the same operator from the beginning to the end.
